Studies on the Pseudomonas phage

Abstract
The properties of Pseudomonas phage isolated from the human stool were investigated and the following results were obtained.
The host range was very narrow and the phage lysed the host bacteria only.
The phage formed two kinds of plaque, large and small. The 2nd generation: of the large plaque phage formed both kinds of plaque, large and small while that of the small plaque-phage formed the small plaque only.
The serological properties of the phages originated from large and small plaques were similar.
When the concentration of host cell and phage was 9.4 x 107/ml and 1.14 x 106 /ml, respectively, about 75% of the phage was adsorbed at 37¡ÆC for 10 minutes.
The minimum latent period, complete phage-liberating time and burst size of phage was 20 min., 62 min. and about 120, respectively.
The neutralization of phage activity by the anti-phage serum obeyed first-order kinetics regardless of the serum concentration.
About 82% of the phage, suspended in broth, was inactivated at 60¡ÆC for 10 minutes and the 10-2M MgSO4 did not give any help to the phage stability.
The phage was relatively stable at pH 6.0~10.0.
The inactivation of phage activity by ultraviolet ray followed first-order kinetics. The phage inactivated by ultraviolet ray formed the multiplicity reactivation,. but no photo reactivation.
The ultraviolet-inactivated phage also killed the host bacteria. On irradiating the same dose of ultraviolet ray; the host killing ability of phage was inactivated at about 1/3 of the rate of plaque forming ability.
